Nelson Bay Road is closed near Newcastle Airport after a crash that injured two people this afternoon.
Paramedics and police were called to the scene about 3.40pm, closing the road in both directions and preparing to airlift one person to hospital.
Northbound traffic is being diverted into Medowie Road, while southbound motorists are heading along Richardson Road.
An Ambulance NSW spokesman said one person had sustained head, chest and leg injuries in the accident.
A second patient was being treated after suffering head injuries, the spokesman said.
A Westpac Rescue Helicopter Service spokesman said a man in his late 40s was being flown to the John Hunter Hospital with multiple injuries.
Crash investigators are also headed to the scene.
The road closure follows an influx of holiday traffic through the region, which began bottlenecking at several points throughout the day.
Queues have been reported at Bulahdelah since 10.30am, stretching to about 5 kilometres at 4.20pm.
Motorists coming off the F3 at Beresfield were also facing delays due to queues of 2.5 kilometres.
